In "Return of the Jedi" - actually many great moments there- Like when Han and Luke are reuinited and brought to Jabba the Hut:
Han: How we doing?
Luke: Same as always.
Han: That bad huh?
Or- when Luke is standing at the end of the plank, over the Sarlack and says "Jabba, this is your last chance- free us, or die." ( I was wondering whqat the heck he planned to do from there-
Or- the very first time I saw Ghostbusters- when I saw the giant Marshmallow man coming down the street I laughed so hard I just about fell out of my seat.
Ace Ventura- when he talks out of his butt- again-I busted a gut.
Lesser known moment maybe- in "O God, You Devil" When Ted Was looks over his wine and says "Hawaiian Chablis?... I've never seen wine with an expiration date before."

How about the moment in Star Trek II: The Wrath of Khan when the rapidly dying Spock in the radiation chamber, facing away from the camera and slowly getting to his feet, reaches down to straighten out his uniform tunic. Just so characteristic.
Another great Trek moment in number VI (The Undiscovered Country -- the specially prepared photon torpedo hits the cloaked Klingon vessel. Captain Sulu leaps to his feet: "Target that explosion and fire!"
Two from Captain Renault in Casalanca: "I'm shocked, shocked to find that gambling is going on in here!" And from the end: "Major Strasser has been shot! (pause) Round up the usual suspects."
(I was just looking over the quotes on the IMDB to get the right wording for those two...damn, that's a great movie. Gotta watch it again soon.)
Actually, I'll submit a third from that film, and possibly my all-time favorite: the singing in Rick's of La Marseillaise.

I second EQT on Casablanca, so many moments.
One of my favorite moments is a bit obscure and old, but I'm a Pacino fan. I love his "opening statement" in ...And Justice for All.

I just watched Before Sunrise for the first time in a few years. The whole movie is brilliant (and makes me hate myself for never backpacking through Europe, but I suppose I could still do that), but Jesse's little soliloquy in which he persuades Celine to get off the train and spend the day with him is just incredible - funny and almost convincing at the same time. "When you're old and married and feeling tied down, you'll remember that all those other guys you knew were a bunch of boring losers like me and you'll figure it's just as well!" Brilliant, I tell you.

The first time we meet Cap'n Jack Sparrow in Pirates, that had me laughing. I had been through a horrible day, and I laughed so hard... probably got weird looks.
I love the part in the Matrix when Neo has defeated Smith and Agent Brown looks at the other and his look says "OH CRAP!" and then they run.
I like the scene in V for Vendetta where V has killed all the rest after being shot at and is still standing, and Creedy screams "Why won't you die?" And V tells him that ideas are bulletproof.
The scene in Watership Down when Hazel rushes to put his plan into action.
Two Towers when the elves march into Helm's Deep, not in the books but I love it. Also in ROTK when the camera travels over the mountains - I'm not sure why but the combination of music and image always make me smile. I also love when we see Eowyn escape from Wormtongue and runs out of the Golden Hall, that is my favorite theme music as well.
Star Wars Episode III when we see the proof before our eyes that Anakin has gone dark. return of the Jedi when Luke does his flip and I thought - aha! That's what he planned... WTF?
The ending of King Kong. Totally cool.
Grave of the Fireflies, when I realized that despite my hopes to the contrary the ending the audience foresaw in the beginning had come to pass. I still cry.
Atticus at the trial in TKAM.
The Lion King when we see the stampede - the animator in me said COOL!
Spirit: Stallion of the Cimmaron. The whole opening scene where the horses are just running I teared up either from how nicely done it was or maybe because I felt the animators's pain...
Life of Brian, the opening scene has be cracking up every time.
Princess Mononoke - The ending I enjoyed, it felt sort of bittesweet.
Castle of Cagliostro - When Lupin and the Inspector lead the cameras down to the counterfeit bills, but the Inspector's acting is SO HORRIBLE the higher-ups are just CRINGEING.
Wow, I am a sucker for shiny cinematography and pretty animation, aren't I?

Oh lord where to start...
- The final courtroom scene in A Few Good Men. Yes I know that scene has been quoted, played, copies, parodied, and knocked off so many times its impossible not to laugh a little bit, but damn the scene was so good. Nicholson was never better and that's saying something. And anyone who says Tom Cruise can't act is wrong. He just chooses not to most of the time.
- The Narrator's "I felt like destroying something beautiful" internal monolgue while he pounds Angle Face in Fight Club.

-When we see the hologram revealing how the reavers came about.
-The Operative and Mal in the temple.
Operative: I already know you will not see reason. Mal: The Alliance wanted to show me reason, they shouldn't have sent an assassin.
Operative: I have a warship in deep orbit, Captain. We locked onto Serenity's pulse beacon the moment you hit atmo. I can speak a word and send a missile to that exact location inside of three minutes. Mal: You do that, you'd best make peace with your dear and fluffy lord. (as he throws a device to Operative)
Operative: ... Pulse beacon
-When Mal gets pissed and decides that they're going into reaver territory.
Mal: This is how it is. Anybody doesn't wanna fly with me any more, this is your port of harbor. There's a lot of fine ways to die. I ain't waiting for the Alliance to choose mine. (shoots an Alliance guy coming out of the ship)

One of my favorite moments is in Lola Rennt (Run Lola Run) when Lola has just robbed her father's bank and bursts out the front door with 100,000 marks in a plastic bag, only to find a SWAT team aiming guns at her. She freezes, thinking that it's all over. Then the cops all start making urgent "get out of the way" motions at her - they don't even know that she's the robber. That has got to be the most hilariously anticlimactic scene ever.

Anyhoo, my contribution to the thread: My favorite scene is when Eminem wins his rap battle at the very end of 8 Mile. After all that he goes through in that movie, choking off the stage, getting beaten up, losing his girl, on and on, he just gets up and destroys the other guy. And then, in total disregard for movie convention, he goes back to finish his shift at the factory. Perfect, perfect scene.

I can't believe no one mentioned the "I am your father" scene in Empire Strikes Back. Best movie moment. Ever.
The Mummy was full of great scenes. The library scene with all the bookcases falling like dominoes is one of the best. And Evie's reaction: "Oops."
ETA: Okay so this is a TV moment, but in Firefly where Wash was playing with the toy dinosaurs: "Mine is an evil laugh."

I just saw this one, but I know it will stay with me for the rest of my life. We rented the Academy Award winning French movie The Chorus and were pleasantly surprised. It was a classic tale of new teacher (or prefect in this case) turns around troubled kids using *****. Like Sister Act 2, ***** was music. Unlike Sister Act 2, this movie was good.
My poignant moment was when the prefect was fired from the school. As he is leaving the grounds, he is disheartened to see that none of his students broke the rules to say goodbye to him. In his words, the children's wisdom came across as indifference. (Before the arrival (and once he left) of the new prefect, punishments for breaking the rules included physical and mental abuse.) As he turns the corner of the building, feeling dejected, he notices small pieces of paper scattered on the ground. As he bends down to pick some up, more pieces fall from above him and he notices that there is writing on them. He looks up to see numerous paper airplanes flying out of the window above him and looking at the ones in his hands, they are goodbye notes from the children. He looks up again and small hands are waving at him from the window as the children begin to sing.
It was so touching to see it played out after all the things he does with the boys and how he is able to change the school employees and their views of the children.

The film itself was long and boring but I absolutely love that massive action sequence in The Matrix Reloaded - the one that starts in the chateau with the swords and armour and things, then moves down into the car park, and then onto the freeway. It was like everything they put in that film - the ghosts, the keymaker, etc - was all to make that one scene possible. It's a delight to see how it all fits together. Not sure if it qualifies as a 'moment' since it's about half an hour long, but whatever.
I also love the scene in the original film when Cypher is unplugging people. It's the dramatic peak of the movie for me.
Also, it's not a film, but that bit in the last episode of Friends where Ross is listening to the answerphone message, and Rachel walks in... it's obvious, but it gives me shivers.

If we're going the Firefly route, I rewatched Serenity last night, and that's got a hundred great moments such as...
...when Simon asks "Am I speaking to Miranda right now?" and River does that incredible "give me a break!" sneer/eyebrow raise.
...when Mal is giving his inspirational speech after finding out the truth about Miranda, and he's backlit so that you can only really see a sunburst as he talks.
..."That's not incense!"
..."Trap?" "Oh yeah, trap."
...the Operative positioning his sword so that the doctor at the beginning can fall on it.
I could go on and on...

The end of Mr. Holland's Opus, in which Glen Holland is being downsized and ready to pack up and leave his lifelong job of being a music teacher, convinced that he's a has-been relic that nobody likes and never amounted to anything.
But his wife brings him to school, where the band, much to Holland's surprise, is waiting. And they play the song that he spent his life writing.

Oh, hell, this is a fun one to think about.
The final scene of "Shawshank Redemption" when the camera cranes back and we see Red walking towards Andy over the beach and hear his voice saying "I hope..." That one always chokes me up, mainly because they don't cheap out and actually show them meeting again, but let us play it out in our imaginations.
The gratuitous "Age of Aquarius" number at the end of "40 Year Old Virgin" surprised me so much I nearly fell off my chair.
The scene in "Office Space" after they have beaten the crap out of the copy machine and are dancing around the apartment to gangsta rap.
I love the "we have such sights to show you" speech from "Hellraiser." Only time a movie monster has ever given me the creeps, he just seems to enjoy it so much.
The sequence in "Evil Dead II" where Ash fights his own hand is brilliant.
I don't know why, but the opening and closing fight scenes from "Blade" just really get me stoked. I always want to put on a long leather coat and go kick some vampire ass after watching it, and at my age that would be ill advised...
The scene in "Dark City" where Bumstead breaks down the wall to Shell Beach startled me when I first saw it. I'll admit it, I never saw that coming, and I love it when a film catches me by surprise.
The "crush your enemies" bit from "Conan" is just great.
The scene from "Raiders of the Lost Ark" where Indy just shoots the guy with the big sword.
The "what's in the box?" bit from "Se7en."
I could go on and on and on and on, but those are a few off the top of my head.

Yes, yes, yes on Serenity . Joss Whedon has an enormous talent for character and dialogue, as do the players he works with. Serenity had so many great moments, in particular the aforementioned scene beginning with the reavers' pursuit of Serenity and ending with the death of Wash, which was most painful. Of all the characters, I think I'll miss him the most.
Other great movie moments that spring to mind:
Life of Brian : "How shall we NSFBSK off, oh Lord?"
The Blues Brothers: After Steven Spielberg gives Jake and Elwood their receipt and they turn to find every gun in the U.S. pointed at them.
A Fish Called Wanda : When Otto holds Archie out of the window by his ankles.
The Empire Strikes Back: The Millenium Falcon navigating the asteroid field. I'll never forget the feeling of seeing that for the first time in the theatre.
Just saw Little Miss Sunshine today, so I won't post any specifics, but it's definitely going on my list of movies with memorable scenes.
